To examine why Band 3 might behave as exhibited in the previous two video problems, Tomishige, et al. treated the interior regions of ghosts very mildly with trypsin, a protease. Under these conditions, the cytoplasmic regions of most Band 3 proteins were digested, but other membrane proteins weren’t affected.

Examine the video results, consider your earlier hypotheses in light of these results, answer the questions and move on to the next problem.

1. Describe the behavior of the particle following trypsin treatment, and compare it with the behavior of a similar particle observed in the first video. You may review the first video in this compartment.
